Rofecoxib - Wikipedia, the free encyclopedia
Rofecoxib (IPA: [rofəˈcɒxɪb]) is a nonsteroidal anti-inflammatory drug (NSAID) that was used in the treatment of osteoarthritis, acute pain conditions, and dysmenorrhoea.
Rofecoxib was one of the most widely used drugs ever to be withdrawn from the market.
Merck has placed great emphasis on these studies on the grounds that they are relatively large (almost 3000 patients) and compared rofecoxib to a placebo rather than to another pain reliever.

Rofecoxib was also associated with improved quality of recovery and increased patient satisfaction.
Rofecoxib is more cost-effective than acetaminophen in preventing pain after ambulatory surgery, reports a study in the October issue of Anesthesiology (2002; 97:931-937).
The new trial supports the use of the selective cyclo-oxygenase-2 inhibitor rofecoxib over acetaminophen for pain control in patients undergoing otolaryngologic surgery.

Vioxx (Rofecoxib) Shows Osteoarthritis Pain Relief Superior to Celebrex (Celecoxib) or Acetaminophen
Rofecoxib 25 mg demonstrated a statistically significant (p less than 0.05) advantage over acetaminophen across every clinical endpoint in the study, including pain from walking, night pain, pain at rest and morning stiffness; celecoxib showed a statistical advantage compared to acetaminophen on only one endpoint (walking pain within first six days).
Efficacy was measured using the Western Ontario and McMaster Universities Osteoarthritis (WOMAC) Index and the Patient and Investigator Global Assessment of Response to Therapy scale.
Statistical analyses were conducted to compare the average response to the different medications after six days and after the six-week duration of the study.

Similar rates of thrombotic events (myocardial infarctions, cerebrovascular accidents, transient ischaemic attacks, angina, deep vein thromboses, and peripheral thromboses) were observed with rofecoxib, placebo, and nonselective NSAIDs (ibuprofen, diclofenac, and nabumetone) in a retrospective analysis of 8 clinical trials consisting of 5435 patients with osteoarthritis.
The above results were supported by a pooled analysis of 23 studies (including the VIGOR study) that represented over 14,000 patients years from patients with osteoarthritis, rheumatoid arthritis, Alzheimer's disease, and chronic low back pain.
Furthermore, similar risk of cardiovascular, haemorrhagic, and unknown deaths, nonfatal myocardial infarctions, and nonfatal strokes was also found between the groups.

By contrast, 26 out of 1,299 patients taking a placebo (2 percent) had had such an event.
