 Lipodystrophy
Lipodystrophies may affect a small area of the body, a large part of the body, or the entire body.
The following conditions may be associated with forms of Total Lipodystrophy: Acanthosis Nigricans (a skin disorder characterized by gray or black darkening and thickening of the skin which occurs frequently in Lipodystrophy patients around the onset of puberty), Acromegaly (gigantism), excessive hair growth (hirsutism or hypertrichosis), eruptive yellowish spots on the skin, and thickened skin.
Centrifugal Lipodystrophy is a rare localized condition characterized by a small skin depression enlarging in a circular pattern.

 Project Inform's Lipodystrophy Syndrome(s) [ HIV / AIDS Treatment Information ]
At various times, lipodystrophy symptoms have been blamed on individual drugs, on classes of drugs, on therapy overall, or on HIV itself.
Since the cause is uncertain, treating lipodystrophy is an inexact science and tries to deal mostly with the changes in body appearance and blood work.
Or, it may even be due to the immune system becoming more aggressive once the onslaught of HIV is slowed down in response to therapy.

Fat Redistribution in HIV Patients - An Opportunity for Drug Manufacturers?
As a consequence, a country-by-country breakdown in the number of patients with lipodystrophy is difficult to determine at present.
As a consequence, patients are requesting changes in drug therapy to combinations that decrease the risk of developing this syndrome.
There is an approximately five-fold increase in the risk of developing lipodystrophy when using combinations of NRTI with PI drugs compared to using each class alone.

As with the management of obesity in the general population, patients and providers are faced with a frustrating inability to affect the debilitating changes in body fat that are observed in individuals on combination antiretroviral therapy.
Until such time as we do have effective treatments for these lipodystrophies, it is very important to work on those variables that can be controlled.

This study is to assess the effectiveness of fish oil when used alone and in combination with sitostanol in lowering the triglycerides (blood lipid levels) and normalizing cholesterol levels in HIV-infected patients on antiviral therapy who have elevated triglycerides.
Patients will be closely evaluated through out the study for the metabolic parameters and body shape changes suggestive of lipodystrophy associated with HIV-infection.
Other trials are directed towards finding newer therapies for patients who have already developed lipodystrophy.

Research provides better understanding of lipodystrophies disorders
Garg has also studied more than 200 families with various types of inherited lipodystrophies and hundreds of patients with HIV who have developed this condition.
Recently, many HIV-infected patients being treated with a class of drugs known as protease inhibitors have developed lipodystrophies, which has raised awareness of these rare disorders.
UT Southwestern researchers have led the way in identifying gene mutations responsible for several forms of lipodystrophies and in identifying novel therapeutic approaches for these patients.

A lipodystrophy can be a lump or small dent in the skin that forms when a person keeps performing injections in the same spot.

Delineation of the human molecular genetic basis of forms of inherited lipodystrophy may help to identify pathways and molecular targets for the common insulin resistance syndrome and for acquired drug-induced lipodystrophy syndromes.
Lipodystrophy syndromes, which include both genetic and acquired forms, resemble the insulin resistance syndrome, particularly with respect to the presence of associated biochemical disturbances such as hyperlipidemia and diabetes.
This may require a better understanding of the mechanism of action of endostatin at the molecular level, which remains largely unknown.
