Amantadine has been used by some HCV patients, but caution must be taken when this drug is prescribed by a medical doctor.
She said the response to treatment might have been greater with a group of patients who had never been treated with interferon, since her study only treated those with interferon- resistant disease."We need support for further studies," said Smith.
Forty percent of the patients achieved a partial response (defined as a reduction in ALT of greater than 50%), and 30% failed to respond to amantadine therapy.

The National Institutes of Health (NIH) Consensus Development Program: Amantadine: Does It Have a Role in the ...
When amantadine hydrochloride is given for prophylaxis, it should be used as adjunctive therapy until the patient has received influenza vaccine and an immune response can be anticipated.
This Consensus Development Conference was called to review the information available on the use of amantadine hydrochloride in the prevention and treatment of disease caused by influenza A. The format utilized was the convening of a panel of individuals with various backgrounds but little or no personal involvement in research on amantadine hydrochloride.
Although amantadine-resistant variants have not as yet been isolated as the predominant virus from patients who have received the drug, the possible selection of such strains in the population under the pressure of extensive amantadine therapy must be considered.

This study does not, of course, answer all of the issues surrounding the use of amantadine as a part of HCV therapy.
Using a multivariate analysis, genotype, pretreatment HCV viral load, age, and amantadine therapy were predictors of sustained virologic response.
During therapy, detectable viremia occurred less often in the amantadine than in the placebo group [12% vs. 22%; P=.04].

Does Amantadine Help Response to HCV Therapy?
In the amantadine triple therapy group response was 38% at 4-6 months of follow-up.
The authors concluded that in this group of previous non-responders, triple amantadine therapy was associated with significant early viral undetectability, and durability is being followed.
I think its important to bear in mind that a number of viral kinetics studies have been conducted using high dose induction therapy, and it is my opinion that it has yet to be firmly established that very early high viral load decline rates produces higher sustained virologic response rates.

Motor-learning Impairment by Amantadine in Healthy Volunteers
In the amantadine group, a further analysis controlling for sex between performance index and amantadine concentrations was carried out, which revealed that the negative correlation shown in Figure 4 was no longer significant.
However, motor-learning accuracy was lower in the amantadine low-performance subgroup than in the amantadine high-performance subgroup, the placebo group and the placebo low-performance subgroup (P<0.01; Table 2), suggesting decreased ability of the amantadine low-performance subgroup to perform under the influence of the drug.
Comparison between amantadine and placebo groups was made using an ANOVA for repeated measures, followed by a contrast analysis (see the text for significance).
